Comments: We have used DRAQ5 for Amnis ImageStream analysis for several years now and it has a nice consistently bright staining. We found this dye to be more reliable and less problematic (bleeding into subsequent samples, contaminating tubes, compensation issues) than various other dyes such as Dapi and hoechst dyes. We add 2-3ul of a 1:1000 dilution to our cells (1-2x10^6)/100ul just before acquisition. Officially our vial has expired (for at least a year), but we have seen no reduction in signal. We have used DRAQ5 for traditional flow-cytometry as well, to either eliminate or select cell-doublets or 2-cell conjugates. For flow we use a 10-fold higher concentration than for imaging, which makes it less cost-effective than some other live dyes in this setting.
