Comments: DRAQ5 is our most established nuclear stain for cell based assays. It also stains the cytoplasm to some degree which is useful for cytoplasm detection. It's emission in the far red is useful as it does not cause cross signals on lower wavelength-excited channels (while Dapi or Hoechst do)! It is quite expensive though! Comments: DRAQ5 is incredibly easy to use. Just dilute and add to cells immediately before acquiring days. Dye incorporates quickly into both live and fixed cells. Comments: Overall DRAQ5 is an excellent product with simple and easy to use instructions.  The dye has very good sensitivity and specificity and can be easily plugged into any protocol with minimal change.  DRAQ5 has cut down on protocol time and the high concentration of the stock solution allows one to conduct multiple studies with one vial of solution; thus dramatically reducing costs and time compared to similar nuclear dyes.

Comments: We have used DRAQ5 for Amnis ImageStream analysis for several years now and it has a nice consistently bright staining. We found this dye to be more reliable and less problematic (bleeding into subsequent samples, contaminating tubes, compensation issues) than various other dyes such as Dapi and hoechst dyes. We add 2-3ul of a 1:1000 dilution to our cells (1-2x10^6)/100ul just before acquisition. Officially our vial has expired (for at least a year), but we have seen no reduction in signal. We have used DRAQ5 for traditional flow-cytometry as well, to either eliminate or select cell-doublets or 2-cell conjugates. For flow we use a 10-fold higher concentration than for imaging, which makes it less cost-effective than some other live dyes in this setting.

Comments: This product is very useful for us in intracellular staining of receptors for localization. It is an easy to use nuclear counter stain because of its advantageous spectral properties (it does not cover at least two or three out of the available visible range optical channels in confocal microscopy, like propidium iodide). Sometimes we can also utilize its unique property that it does not need cell permeabilization that other nucleic acid stains require.
