Comments: I’ve been really pleased with DRAQ5 and have found it both reliable and easy to work with. I’ve used it for flow cytometry and immunofluorescence microscopy, and even when detecting very small amounts of DNA (in platelets!) it was sensitive enough to give clear, consistent results. It’s straightforward to use, works well across different applications, and is also very reasonably priced - overall, a great reagent that I’d highly recommend.

Comments: To test the effects APPA (10 µg/ml), AP (2.3 and 10 µg/ml) and PA (7.7 and 10 µg/ml) on T/C28a2 chondrocyte viability, DRAQ7TM (Thermo Fisher Scientific, Waltham, MA, USA) was used. Following 24 hours (h) of different treatments, cells were trypsinized and centrifuged at 1,500 rpm for 10 minutes (min). The harvested cells were suspended in 3 µM of the fluorescent dye DRAQ7TM (Thermo Fisher) and incubated in the dark, at room temperature (rt) for 10 min. The samples were run on CytoFLeX flow cytometer (Beckman Coulter Inc. Pasadena, Ca, USA). At least 1x104 cells per assay were measured. Data were analyzed using CyExpert V 2.5 software (Beckman Coulter Inc). Positive control cells were treated with 5 µM Eto plus 10 ng/µl OSM while negative control (Basal) represented cells without treatment.

Comments: We used Invitrogen™ eBioscience™ DRAQ5™ primarily as a far-red nuclear stain for both flow cytometry (FACS) and confocal microscopy applications. In flow cytometry, DRAQ5 provided rapid and uniform nuclear staining, enabling accurate discrimination of nucleated cells from debris. Its far-red emission allowed us to combine it with commonly used fluorophores such as FITC, PE, and APC, without significant spectral overlap, thus expanding our multicolor panel design.

For cell sorting, DRAQ5 proved particularly useful in identifying nucleated versus enucleated cells and facilitated precise gating strategies. The dye’s rapid uptake, even without permeabilization, made it highly suitable for live-cell analysis and sorting with minimal sample preparation.

In confocal microscopy, DRAQ5 provided sharp, high-contrast nuclear labeling with minimal photobleaching. Its far-red excitation minimized phototoxicity and avoided interference with green and red fluorophores, allowing us to perform multiplexed imaging of protein markers in both fixed and live-cell conditions.

Overall, DRAQ5 performed robustly across both platforms, delivering reproducible results and consistent staining quality. We selected it for its spectral properties, ease of integration into FACS workflows, and compatibility with confocal imaging, and it has become a reliable part of our staining toolkit.

Comments:                 DRAQ9 was a gift from BioStatus. We used it to determine if the vacuoles that we were observing in mammalian cells under hypotonic stress are surrounded by a lipid membrane. DiI that we tried before failed to stain the vacuoles, but with DRAQ9 the result was unequivocally positive: a bright staining was easily detectable under a confocal microscope. The presence of lipid was confirmed with Nile Red. We published the image in Fig. 5 of https://www.nature.com/articles/s41598-024-79815-z.pdf

Comments:  I use DRAQ7 for cell staining to enable a precise and reliable quantification of the viability of my cells before and after the application of compounds at varying concentrations. I have never had any problems with the reproducibility of results, with clear and precise control values every single time. There is honestly nothing bad I can write about this product as I have never had any problems, and it's so simple to use. Our main problem with these stains is that they cannot be used in any microscope, e.g. Draq5 can be used in a confocal laser microscope, but cannot be used in another, simpler microscope, so we have to purchase different stains for different microscopes. In addition, DraQ5 is more expensive than other similar stains.

Comments: The DraQ5 fluorescent probe is supplied as a water-soluble concentrated solution (0.5mM or 5mM) that can be diluted depending on the desired application. Given its high affinity for double-stranded DNA, DraQ5 can be used as a nuclear counterstain in immunohistochemistry studies upon short incubation periods of time, as an alternative to blue-fluorescent DNA-binding probes. However, the broad excitation and emission peaks must be taken into account if a combination with other fluorophores is desired. In our experience, it is also compatible with aqueous-based tissue clearing techniques, rendering a uniform labelling across entire mouse brain hemispheres (55-days-old) that could be imaged by light-sheet fluorescence microscopy. To achieve this, a simple incubation at 37⁰C/60rpm for 12h with a DraQ5 diluted solution (2.5uM) and a 12-hour washing step at 4⁰C to remove excess were incorporated in the tissue clearing procedure.

Comments:                 Without Cygel it would be almost impossible to obtain high-quality microscopy of motile forms of Leishmania parasites. The product is easy to use and gives a reproducible performance. I've used it to image Leishmania promastigotes that were modified to contain fluorescent proteins, in order to gain information on protein function. Cellular morphology remained unchanged by the embedding process and protein localization was not affected. Can highly recommend Cygel.

If staining is not stable over time that can be the case in live cells, where after several hours a re-distribution can occur in some cell types, mostly towards lipophilic structures in the cytoplasm (Martin, 2005). If the staining is not stable in fixed cells then this may be due to washing post-staining, too low concentration, or not removing surfactants used in prior cell permeabilisation procedures.
