Comments: Good nuclear marker for single- and double-labelling immunofluorescence and confocal microscopy on frozen and paraffin-embedded sections. Staining disappears in sections pre-treatment with formic acid (as in prion diseases). Sometimes, non-specific staining of other structures.

Manufacturer's response: Thank you very much for the valuable feedback and product review on DRAQ5. The observation about the use of formic acid is useful to know. Perhaps it may be worthwhile to attempt staining with DRAQ5 as the final step after all other steps are complete, including the formic acid treatment. It is unusual to see staining of other structures outside of the nucleus with DRAQ5 but may be a consequence of the treatments of the sample. DRAQ5 does have, like all cell permeant drug-like small molecules, a propensity for binding to lipophilic structures in addition to the specific target, in its case dsDNA, but this is usually only seen in cases of understaining, paradoxically. We are always ready to answer these questions through our tech support service as they occur in real-time in the bench experiments.
