Comments:                 We used CyGel for imaging a K562 cell line to verify that our fluorophore-tagged biosensor is internalized by the cells. The product was very easy to use and worked according to the manufacturer’s description. There was no problem with CyGel interfering with our fluorescent signal using a standard fluorescent microscope for cell imaging. We think it was a very reasonable price for the ease of use and results we obtained. While we did not require follow up service, it was there for our assistance as needed.

Comments: DRAQ7 was used to measure cell death in complement-dependent-cytotoxicity experiment.  
Our project was to determine the ability of anti-Tn antibody to mediate death of Tn expressing in association with complement. Briefly, Tn expressing cells were incubated with anti-Tn antibody, in the presence of complement and DRAQ7 as a viability marker. Cell were incubated in Incucyte System (www.essenbioscience.com/en/products/incucyte?gclid=CObfsKOZjM8CFWMW0wodVw0JSQ) and the appearance of red fluorescence (staining of dead cells by DRAQ7) was monitored every 30 minutes.  We were able to measure cell death over time by the use of DRAQ7. Fluorescence emission signal was high, and the results obtained were reproducible.

Among all viability dye we have tested, it was the only one that emits strong signal after several hours of incubation.

Comments:                 Imaging mitochondrial dynamics requires that my sample be completely stationary and alive! CyGel allowed me to stabilize zebrafish larva and obtain time-lapse images of mitochondria. I was able to rescue the fish afterwards, so that fish could be imaged on multiple days if necessary.

Comments: Worked with diluted stock solution in PBS (better handling and accuracy). The applied incubation time, temperature and concentration were important for good reproducibility. On permeabilized cells the nucleus had better signal and stained clearly.  On the non-permeabilized cells, the cytosol was stained also.

Comments: I use DRAQ5 in order to separate living cells of dead cells in a suspension of brain tumor cells and to calculate the cellular DNA content. I use 3 ul of DRAQ5 3 minutes before the acquisition, therefore I spend several vials of DRAQ5 in a short time and it is an expensive dye.

Comments: DRAQ5 was used to visualize nuclei in live HeLa cells using confocal fluorescence microscope LSM 510. This product is very stable and convenient to use. The protocol involving dilution and staining is very simple and easy to follow. The dye is not toxic to HeLa cells and bio-safe. The dye did not interfered with subsequent downstream biochemical analysis e.g. western blot and protein band detection by LI-COR.  DRAQ5 uses far red wavelength, it does not involve the risks associated with UV light as associated with some other nuclear staining dyes in market. Also it was advantageous to us since nuclei visualization was the most essential part of our experiments and DRAQ5 imaging was compatible with our LSM 510.

Comments: We used DRAQ5 as nuclear staining in Flow Cytometry and ImageStream analysis to track the subcellular localization of different proteins. It works well with different detectors and offers a nice alternative to DAPI, leaving UV fluorophores available for other markers. 

This product is also very convenient in tissue immunofluorescence as a cellular marker (nuclei). It allows easy characterization of immune infiltrates in diverse tissues with better and more consistent signal than DAPI.

Comments: After assay optimization, this product worked quite well for imaging host cells by microscopy during bacterial infection. The product has a long shelf life, provided consistent results, and was not that expensive. The only reason I gave 4 starts for the After Sales Care is because I had no direct interaction with them, but I think our lab tech found the initial instructions with the package confusing. Overall, it is a great product for a specific assay.
