Manufacturer's response: Thank you very much for the valuable feedback and product review on DRAQ5. The observation about the use of formic acid is useful to know. Perhaps it may be worthwhile to attempt staining with DRAQ5 as the final step after all other steps are complete, including the formic acid treatment. It is unusual to see staining of other structures outside of the nucleus with DRAQ5 but may be a consequence of the treatments of the sample. DRAQ5 does have, like all cell permeant drug-like small molecules, a propensity for binding to lipophilic structures in addition to the specific target, in its case dsDNA, but this is usually only seen in cases of understaining, paradoxically. It is indeed a well-understood phenomenon that all cell-permeant DNA-binding probes will have some impact on cell behaviour. Accordingly, we alert users to this on the front of the product’s technical datasheet. Nonetheless, DRAQ5 has been widely used for live-cell end-point assays and as a means of identifying the nucleated cells in flow cytometry experiments on complex sample matrices such as bone marrow, besides its popularity as a standard IF counterstain.

Manufacturer's response: Thank you for your review of DRAQ5. DRAQ5 does have a useful weak secondary staining of the cytoplasm which HCS labs use to segment the cell envelope, by increasing the gain. An unwanted inversion staining (“hollow” nuclei) is a consequence of too low a concentration of DRAQ5. We strongly recommend 5 uM for fixed cells and 5-10 uM for live cells. We would be very happy to discuss this and troubleshoot the issue to try to resolve it.
