It is indeed a well-understood phenomenon that all cell-permeant DNA-binding probes will have some impact on cell behaviour. Accordingly, we alert users to this on the front of the product’s technical datasheet. Nonetheless, DRAQ5 has been widely used for live-cell end-point assays and as a means of identifying the nucleated cells in flow cytometry experiments on complex sample matrices such as bone marrow, besides its popularity as a standard IF counterstain.

Manufacturer's response: Thank you for your review of DRAQ5. DRAQ5 does have a useful weak secondary staining of the cytoplasm which HCS labs use to segment the cell envelope, by increasing the gain. An unwanted inversion staining (“hollow” nuclei) is a consequence of too low a concentration of DRAQ5. We strongly recommend 5 uM for fixed cells and 5-10 uM for live cells. We would be very happy to discuss this and troubleshoot the issue to try to resolve it.

Manufacturer's response: The initial review posting elicited an immediate technical support response. After successful testing of both fresh DRAQ5 and DRAQfx (for comparison) in the customer’s lab the issue was isolated to the presence of 0.3% Triton X-100 during the final counterstaining step. This was remedied by an intermediate wash with PBS. This learning will be reflected in our products’ technical data sheets for fixed cell immunofluorescence protocols.

Manufacturer’s response: The presence of precipitates in the vial is most unusual and could only be a result of contamination or freezing (accidental, most usually). We would like to have explored this with you at the time you saw this. Your comment about smaller quantities for fixed cell staining is well made - we have developed a product exactly of this nature - please see DRAQfx which is specifically for fixed cells, provided in a convenient dropper bottle. <A replacement vial of DRAQ5 was provided free-of-charge>.

Manufacturer's response: Thank you Anirban! Yes, DRAQ5 will also work for fixed cell fluorescence microscopy. You can use it prior to fixation, but more usually it is used AFTER fixation of the sample. We also have a new convenience alternative - DRAQfx FIX & GO - which has been specifically designed for the latter use and may exhibit less cytoplasmic background, though typically this is extremely low.
