You make a good point too: it is possible to view DRAQ5-positive cells with transmission light microscopy - the nuclei are indeed deep blue (as well as far-red fluorescent)! It’s a convenient way to test for functionality and sample quality (for example prior to FACS) and might even allow for a new breed of assays.

The product code is DR05500 and, having reviewed your protocol, would perhaps allow you to skip the pre-dilution step entirely e.g. 4 µl into a final volume of 100 µl would give a final concentration of 20 µM. Perhaps this might assist you better?

Manufacturer's response: Thank you very much for the valuable feedback and product review on DRAQ5. The observation about the use of formic acid is useful to know. Perhaps it may be worthwhile to attempt staining with DRAQ5 as the final step after all other steps are complete, including the formic acid treatment. It is unusual to see staining of other structures outside of the nucleus with DRAQ5 but may be a consequence of the treatments of the sample. DRAQ5 does have, like all cell permeant drug-like small molecules, a propensity for binding to lipophilic structures in addition to the specific target, in its case dsDNA, but this is usually only seen in cases of understaining, paradoxically. We are always ready to answer these questions through our tech support service as they occur in real-time in the bench experiments.

It is indeed a well-understood phenomenon that all cell-permeant DNA-binding probes will have some impact on cell behaviour. Accordingly, we alert users to this on the front of the product’s technical datasheet. Nonetheless, DRAQ5 has been widely used for live-cell end-point assays and as a means of identifying the nucleated cells in flow cytometry experiments on complex sample matrices such as bone marrow, besides its popularity as a standard IF counterstain.
