Manufacturer's response: Thank you for your review of DRAQ5. DRAQ5 does have a useful weak secondary staining of the cytoplasm which HCS labs use to segment the cell envelope, by increasing the gain. An unwanted inversion staining (“hollow” nuclei) is a consequence of too low a concentration of DRAQ5. We strongly recommend 5 uM for fixed cells and 5-10 uM for live cells. We would be very happy to discuss this and troubleshoot the issue to try to resolve it.

Comments: DRAQ7 is a very stable and easy-to-use dye. We carefully titrated dye using fix/perm approach after the staining for 7 surface markers. After optimization of the staining protocol we achieved strong signal with an easy discrimination of nucleated cell from the cellular debris and fat particles. Samples were analysed on Navios flow cytometer.

Comments: This product allowed us to effectively differentiate live and dead cells and debris when used in concert with propidium iodide for flow cytometric analysis. DRAQ5 can be diluted directly in FACSFlow when resuspending cells and offers a short incubation time. We were able to use this DRAQ5 when analyzing viability in several cell treatment groups and the results obtained matched the expected results.

Comments: We used DRAQ7 to monitor the cell death along implants over time. For this purpose we required a non-toxic and stable fluorescent dye to detect dead cells. DRAQ7 met all necessary requirements and worked also in the hydrogel. It was very easy to use with immediate results without optimization period.  Some other advantages are that it works without UV light and is non-toxic to cells over time.

After successful testing of both fresh DRAQ5 and DRAQfx (for comparison) in the customer’s lab the issue was isolated to the presence of 0.3% Triton X-100 during the final counterstaining step. This was remedied by an intermediate wash with PBS. This learning will be reflected in our products’ technical data sheets for fixed cell immunofluorescence protocols.

Comments: The product is very easy to use and I followed the manufacturer's instructions. It works very well and does staind the nuclei. Unfortunately 6 months after opening the quality got lower since precipitates were shown in my images. I kept it at 4° as requested. It would be useful to be able to buy smaller aliquots for labs only using it to stained fixed cells.

Comments: My colleagues and I could see a clear and bright signal even with low laser power. There was never an issue of poor dye penetration into 3D cellular aggregates, as would otherwise be observed with more common nuclear dyes, such as Hoechst . The quality of staining is usually consistent across experiments  and among different colleagues. This follows that our results were also consistent across experiments. The dye was stable after repeated uses, and the signal intensity of DRAQ7 did not decrease with repeated use. Moreover, as it is simply added to cell culture media, it fits easily into any experimental design.  I would recommend DRAQ7 to other researchers for its stability, fluorescence quality and its convenience of use. One suggestion for improvement would be to create versions in other fluorescence colors instead of just one which is presently APC.

Comments: We used DRAQ7 to visualize the formation of a necrotic core in fresh isolated Islets of Langerhans, which are prone to hypoxia.  DRAQ7 was simply added to the medium to a final conc. of 1 microM. This compound appeared specific for staining the nuclei of necrotic cells only. It is easy in use, provides a fairly low background and stable regarding bleaching.

Comments: DRAQ5 stain nuclei in an easy way, I have no doubt that is one of the best, mainly in live cells, though for the specific case of my lab we rather used cell-fixed preparations, though for this case DRAQ5 works fine as well. Comments: DRAQ5 from BioStatus is a very useful reagent for cytometry-related applications. For our study, it allowed us to discriminate between nucleated cells and anucleate cells. In addition, within the anucleate cells, it allowed us to discriminate between erythrocytes and reticulocytes. This reagent is very easy to use, extremely reliable and efficient as it quickly enters cells. It displays a very bright fluorescence with only a very small amount. Support from BioStatus is outstanding. It is also very easy to use and does not need to be washed.

Comments: The product works very well and can be re-used. I used it as an alternative to DAPI and Hoechst, to stain cellular DNA within the nucleus, because the CLSM system we use does not have a 405 nm laser line. The main problem of this product is that it is very expensive when compared to other stains such as the ones mentioned above. Indeed for such application I would not recommend it. However, it is ideal for live cell imaging experiments since it is cell permeable and the 633 laser used for its excitation is much less phototoxic as compared to the 405 one.

Manufacturer's response: Dear Gualtiero, Thanks for your considered review of DRAQ5 and we note your comment on cost. DRAQ5 is second generation DNA probe that enables live or fixed cell counterstaining and avoids UV exposure as you rightly observe, plus with much lower photo-bleaching and remarkable shelf stability. Being red-excited it also enables use of the new violet-excited chromophores to extend channel capacity. Bearing these significant benefits in mind, the cost per slide of around 30 eurocents (based on 5uM, 200ul per slide, 200ul pack list price) would seem to be not unreasonable!
