Comments: DRAQ5 is a useful reagent for analysing the cell cycle status of live/dead cells by flow cytometry. For our purposes, cells were first killed by fixation in 70% Ethanol  (because we were co-staining for other antigens) and eventually resuspended in buffer containing 2ul/ml DRAQ5 allowing 10 mins prior to flow cytometric analysis. Lovely DNA profiles were obtained with no compensation issues with the other fluorochrome (PE) used in the study. Never attempted on live cells but we understand from other colleagues in the department that this is possible. Care must be taken when handling as this is a toxic substance.

Comments: Effective and easy to use, the signal could be weak sometimes, but this could have been experimental variation or the microscope itself. Had no problem being able to pseudo-colour and the images and contrast with other stains was generally very good. The sales/technical team were very good before purchase, after-sales care not really applicable, as we didn't need to get in touch after buying it.

Comments: DRAQ5 is great as it can be used to stain living cells as well as fixed ones.  Of course, the cells won't  divide in it, so you can't use it for long-term time-lapse imaging, but it's still very useful for shorter time lapses.

This is important information – we do not recommend DRAQ5 for long-term exposure to live cells as it, like ALL high affinity DNA binders and intercalators, will have some kind of impact on cell health. There are exceptions especially with terminally-differentiated cells as observed with HUVEC, macrophages and BaF3. However, short-term exposure and use in live-endpoint assays has been a hallmark of the product’s success in drug discovery and flow cytometry applications, in particular the loss of cytoplasmic GFP signals when cells are partially permeabilized by fixation with formaldehyde and the unspecific loss of cells due to RBC lysis and associated washes, respectively.

Journal of the American Heart Association. 2017 6(4)). We labelled the cells with 1 uM DRAQ5 for 10 minutes prior to imaging, which was lower than the recommended concentration, but it fluoresced nicely.

Comments: The dye was highly effective as even at very diluted concentrations produced excellent results. The confocal images obtained from these chondrocytes whose nuclei were labelled with DRAQ5 produced high quality and reproducible results. I had used this dye to count the number of cells in clusters because in my culture experiments the cell membranes were frequently touching and therefore thresholding of individual cells was impossible. In order to count number of cells in clusters I counted number of nuclei and DRAQ5 gave me highly reliable results as I could count the number of nuclei in 3D by using Volocity (a 3D image analysis software). It is quite an expensive dye but as the very diluted concentrations are very effective therefore, justifies the cost. Moreover, it was easy to use and highly penetrable and required very little time to label nuclei efficiently.

Comments: The dye gives strong staining and could we washed away even from live cells. Nuclei of some cell lines e.g. PC3 could not be stained (the dye does not enter the cells) others e.g H1299 contain strange dotty staining in the cytosol in addition to the nuclear staining. Too high concentration causes artificial condensation of chromatin.

Comments:                 For many years, my core facility users in general and I, personally, have been using several BioStatus products, mainly DRAQ5 and DRAQ7. I have always found them effective and extremely easy to use. DRAQ7 has shown for many years to be a fantastic option for the detection and exclusion of dead cells. Quite recently, we have tested and used DRAQ7 DROP & GO for the detection and exclusion of dead cells from a very heterogeneous sample for isolating photoreceptors from adult zebrafish retinae for downstream RT-PCR. It was as easy as described in the name of the product "drop and go". Dead cells were clearly differentiated from the alive ones as can be easily observed in our recently published paper Glaviano et al. "A method for isolation of cone photoreceptors from adult zebrafish retinae" BMC Neuroscience (2016) 17: 71 (DOI 10.1186/s12868-016-0307-2)

Comments: Product (DRAQ5) nucleic acid stain works exceptionally well, and keeps common microscope filter channels open since it's a far red stain. Little to no background, easy to use and store. Penetrates deep into thick tissues so you don't observe staining concentration gradients as with other nuclear stains. Comments: If you are looking for a dye for in vivo staining, DRAQ5 is the best. Even with very little amount of a stock of the product, like 1ul, you can observe bright nuclei immediately. Staining genomic DNA specifically you could capture where DNAs are distributed in nuclei. After decorating your nuclei with it, cells are still happy living healthy. I will keep using the product for my future projects.
